Protein characterization

  • Biochemical characterization (enzymology, kinetics, sequencing, protein-protein interactions, proteomics, etc)
  • Biophysical charaterization (microcalorimetry, dynamic/static light scattering, protein stability, folding & aggregation, X-ray crystallography, etc)
  • Our Robotein Platform encompasses an Octet system that allows high-throughput analyses of protein interactions in 96- and 384-well microplates. It enables real-time and label-free determination of affinity, kinetics and concentration. This makes it highly suitable for the characterization of drug candidates. Rapid screening and epitope binding assays are enabled by the multiplexed biosensor formats

Protein production & purification

  • Genetic engineering (site-directed mutagenesis, random mutagenesis, protein design) and cloning in bacterial strains such as Coli, Bacillus subtilis, Bacillus amyloquefaciens, Streptomyces lividans or in yeast such as Pichia pastoris
  • Analytical scale or pilot scale recombinant protein production in high cell density conditions
  • Cell harvesting or supernatant cleaning using bucket or continuous centrifugation or hollow fiber filtration
  • Cell disintegration to recover proteins produced in the intracellular compartment
  • Protein purification at analytical and pilot scales

 

  • Microplate shaker (6 microtiter or Deepwell plates from 6 to 96 sample wells, 300 µL to 3 mL working volumes adapted for strain screening, mutagenesis analysis and multiple parallel culture conditions monitoring (culture medium, induction)
  • Incubator shaker: large capacity Innova 44 and refrigerated Innova 43R (Eppendorf) programmable incubator shakers that accommodate numerous culture flasks are available for culture volumes of 10 mL to 1 L and working temperatures from 4°C to 80 °C
  • Computer controlled fermentors (working volume from 1 to 80 L for batch and fed-batch cultures
  • Cell disruptor (Emulsiflex C3 (Avestin) (7 mL to 300 mL) or our NS 1001L Panda 2K (GEA Process Engineering Division) (> 200 mL))
  • Systems for semi- or totally automated purification of proteins including: ÄKTA Prime, ÄKTA Purifier 100, ÄKTA Explorer 100, ÄKTA Explorer 10S (2D), Biopilot, profinia protein purification system
